Immunoprecipitation:GMDS AntibodyNBP3-29570] -Detection of human GMDS by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-GMDS antibody NBP3-29570 (lot NBP3-29570-1) used for IP at 6 µg per reaction. GMDS was also immunoprecipitated by rabbit anti-GMDS antibody. For blotting immunoprecipitated GMDS, NBP3-29570 was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background

GDP-mannose 4,6-dehydratase (GMD; EC 4.2.1.47) catalyzes the conversion of GDP-mannose to GDP-4-keto-6-deoxymannose,the first step in the synthesis of GDP-fucose from GDP-mannose, using NADP+ as a cofactor. The second and third stepsof the pathway are catalyzed by a single enzyme, GDP-keto-6-deoxymannose 3,5-epimerase, 4-reductase, designated FX inhumans (MIM 137020).(supplied by OMIM)
