Background

S100A12 (Calgranulin C, MRP6, or EN-RAGE) is a calcium-binding pro-inflammatory protein that belongs to S100 family of proteins (family named so as its members are 100% solubile in ammonium sulfate at normal pH) and its expression is restricted to neutrophil granulocytes and monocytes. In normal individuals, it is expresses in tissue/organs where neutrophils and monocytes/macrophages are common, such as spleen and lung, wherein, it is located predominantly in granulocyte cytosol but translocates to membrane as well as cytoskeletal components following interactions with calcium. S100A12 involves in recruitment of leukocytes, promotion of cytokine/chemokine production, and regulation of leukocyte adhesion as well as migration. It acts as an alarmin or DAMP (danger associated molecular pattern) molecule for stimulation of innate immune cells via binding to receptor for AGER that follows activation of MAPK and NFkB signaling leading to production of proinflammatory cytokines and up-regulation of cell adhesion molecules ICAM1/VCAM1. Additionally, it been suggested to inhibit matrix metalloproteinases activity (of MMP2, MMP3 and MMP9 by chelating Zn2+ from their active sites) and also to acts as a monocyte/mast cell chemoattractant, and involve in triggering mast cell degranulation as well as activation that generates chemokines, histamine and cytokines inducing further leukocyte recruitment to the sites of inflammation.
