Western Blot: EDNRA/Endothelin R Type A Antibody [NBP1-33614] - Various tissue extracts (50 ug) were separated by 10% SDS-PAGE, and the membrane was blotted with Endothelin A Receptor antibody [C3], C-term diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (NBP2-19301) was used to detect the primary antibody.

Background

Endothelin A receptor binds endothelins, induces intracellular calcium flux and arachidonic acid accumulation. It is a member of the G protein coupled receptor family. Endothelin A receptor has been reported in a variety of tissues including placenta, uterus, testis, heart, adrenal gland, lung, kidney, ovary, skin, liver, bladder, large blood vessels, cerebellum, neurons, and meningioma tumor cells.

I'm interested in your antibody NBP1-33614, for the detection of the endothelin receptor A (ENDRA) with Western Blot in multiple myeloma cell lines. a) Firstly, I would very much appreciate any feedback from you (i.e protocols, bibliography) of use/applicability of these antibodies for the detection of ENDRA by Western blot analysis in human cancer cell lines and especially in human multiple myeloma cell lines, or in any other human cancer cell line of hemopoietic origin (i.e leukemia). b)Furthermore, is it possible to use the human prostate cancer cell lines PC-3 and LNCaP as positive controls for the detection of ENDRA by Western blot and if it is so I would be very grateful if you suggest me the corresponding bibliography.
   • a) Unfortunately we do not have any information on the detection of ENDRA by Western Blot analysis in cancer cell lines. We also do not have a specific protocol for this antibody as it was tested by an outside lab. The recommended dilution is 1:500 to 1:3000. If you have trouble with this antibody I can reassure you that we have a very dedicated lab team who are always willing to help. b) As a positive control I would recommend NBL1-10117 or one of these tissues: aorta, cerebellum, lung, atrium and cerebral cortex. The protein is most highly expressed in these tissues.
