Background
Principal Names: Cyclin-dependent kinase 5 Official Gene Symbol: CDK5 Gene ID: 1020 (Human) Gene Map Locus: 7q36 (Human). Cdk5 is a proline-directed serine/threonine kinase, belonging to the cyclin-dependant kinase family. It is primarily expressed in terminally differentiated cells, such as neurons, muscle and len fibres. Association of Cdk5 with a neuron-specific regulatory subunit, p35, is required for its kinase activation. The complex of Cdk5 and p35, plays important roles in brain development, such as neuronal migration and neurite outgrowth. The protein is a downstream effector of Rac, a member of the Rho family of small GTPases, which regulates neuronal growth cone motility and the cytoskeletal rearrangements involved in cell migration and ruffling. It also plays a role in regulating both cell-matrix and cell-cell interactions in the non-neuronal cells like len fibres. The pathological role of Cdk5 has been observed in Alzheimer's disease where it promotes neurodegeneration by aberrant hyperphosphorylation of Tau protein.
