Background

Casein kinase I (also designated CKI) and casein kinase II (CKII) compose a family of serine/threonine protein kinases which are present in all eukaryotes examined to date. Casein kinase I family members, which include casein kinase Ialpha, Igamma, Idelta and Iepisilon, have been implicated in the control of cytoplasmic and nuclear processes, including DNA replication and repair. CKII is usually expressed as a tetrameric complex consisting of either an alpha2beta2 or an alphaalpha'beta2 structure. The a catalytic subunit is stimulated by the beta regulatory subunit, which undergoes autophosphorylation. Casein kinase II activity is high in the cytosol and nucleus of proliferating and differentiating cells. Casein kinase II is known to phosphorylate more than 100 different substrates including nuclear oncoproteins, transcription factors and enzymes involved in DNA metabolism.

1. Regarding Casein Kinase 1 alpha Antibody NBP2-50030: 1. The datasheet says the immunogen is recombinant full length Casein Kinase 1 alpha (shortest isoform) [UniProt# P48729], but why does the specificity say this antibody will bind to all Casein Kinase 1 isoforms. Is this a typo? I need the antibody specific to Casein Kinase 1 alpha but not Casein Kinase 1.2. The IHC application suggests a 1:5000 dilution, could I dilute 1:50 for storage? If yes, how long could it be stored?
   • The CK1a gene has two sites of alternate splicing which may or may not insert two sequences called L (long, 28 amino acids) and S (short, 12 amino acids). So there are four possible and actual expressed forms, CK1a, CK1aL, CK1aS and CK1aLS. With collaboration, the lab showed that the L peptide contained a nuclear localization sequence and, using GFP fusions, CK1a forms with L go into the nucleus and those without do not. These are the isoforms mentioned. However, cross reactivity with other isoforms of Casein Kinase 1 such as beta, gamma, delta, or epsilon has not been determined, and so we cannot say for sure whether cross reactivity will occur.
