Background

In cardiac tissue brain natriuretic peptide (BNP) is synthesized as 134 amino acid precursor (prepro-BNP), which is cleaved by proteases to form a 26 aa signal peptide and a 108 aa Pro-BNP (aa 27-134). Proteolytic digestion of Pro-BNP results in formation of 76 aa amino-terminal NT-proBNP (aa 27-102) and biologically active 32 aa BNP hormone molecule (aa 103-134). Both proBNP and NTpro-BNP circulate in human plasma and have been proposed as markers for early diagnosis of left ventricular dysfunction as well as prognostic markers of possible cardiac complications at patients with heart failure.

Granzyme A was detected in immersion fixed human peripheral blood mononuclear cells (PBMCs) using Goat Anti-Human Granzyme A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2905) at 15 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (yellow; Catalog # NL001) and counterstained with DAPI (blue). Recombinant Human Granzyme A protein was serially diluted 2-fold and captured by Mouse Anti-Human Granzyme A Monoclonal Antibody (Catalog # MAB29052) coated on a Clear Polystyrene Microplate (Catalog # DY990). Goat Anti-Human Granzyme A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2905) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998) followed by Substrate Solution (Catalog # DY999) and stopping the enzymatic reaction with Stop Solution (Catalog # DY994).
Western blot shows lysates of NK‑92 human natural killer lymphoma cell line. Immunocytochemistry/Immunofluorescence: Serpin A8/Angiotensinogen Antibody [NBP1-30027] - HepG2 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-Angiotensinogen at 10 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
