active/cleaved Caspase-3 Assay Kit (Fluorometric) Summary

Description
Activation of caspases plays a central role in apoptosis. The Active Caspase-3 Staining Kit provides a convenient means for detecting activated caspase-3 in living cells. The assay utilizes the caspase-3 inhibitor DEVD-FMK conjugated to sulfo-rhodamine (Red-DEVD-FMK) as the fluorescent in situ marker. Red-DEVD-FMK is cell permeable, nontoxic, and irreversibly binds to activated caspase-3 in apoptotic cells.
Detection method: Flow cytometry (FL-2 channel), fluorescence microscopy, fluorescence plate reader (Ex/Em = 540/570 nm).Applications: Detection of activated caspase-3 in living cells.

Background

Caspase 3 (CASP3) is a member of the cysteine-aspartic acid protease (caspase) family that is involved in the execution-phase of cell apoptosis.

There are two classes of caspases involved in apoptosis; initiators (activation by receptor cluster) and effectors (activation by mitochondrial permeability transition). Caspase 3 is considered an effector, as it is activated in the apoptotic cell both by extrinsic (death ligand) and intrinsic (mitochondrial) pathways. Specifically, procaspase is activated when the procaspase is cleaved at a particular residue. Activated Caspase 3 then cleaves numerous substrates to effect the morphological changes associated with apoptosis.

Caspase 3 antibodies are important tools for apoptosis studies and research on the caspase family mediated apoptosis signaling pathway.
