ATP-binding cassette transporter 1 (ABCA1), also known as cholesterol efflux regulatory protein (CERP), is a member of the human transporter sub-family ABCA. ABCA-1 mediates both the export of cholesterol and phospholipids from the cell, as well as the transport of lipids between the Golgi and cell membrane. Specifically, ABCA1 controls the efflux of cholesterol and phospholipids to lipid-poor apolipoproteins, which can then proceed to form new high-density lipoproteins. Regulation of the cholesterol flux between HDL and macrophages is a competitive process between ABCA1 and SR-BI. ABCA1 is expressed on the plasma membrane and the Golgi complex, and is regulated by cholesterol flux. Mutations in the ABCA1 gene have been linked toTangier disease, an autosomal recessive disorder characterized by an absence of plasma HDL, cholesterol ester depositing in the reticulo-endothelial system and deviations in cellular lipid trafficking. ABCA1 antibodies are useful for studies of cholesterol formation and lipid processing mechanisms.
