ABCA1 - The Caretaker for Cholesterol Transportation

Mon, 08/04/2014 - 15:41

The ATP-binding cassette transporter A1 (ABCA1) protein is a key gatekeeper for regulating intracellular cholesterol transport. It is one member of a large family of genes comprised of cAMP-dependent anion transporter cell membrane proteins. These important proteins regulate reverse cholesterol efflux from cells into the peripheral tissues via apolipoprotein A-1 (apo). ABCA1 in particular has a diverse expression profile and is most highly expressed in macrophages. Sporstol's group performed real-time RT-PCR and immunoblotting with the ABCA1 antibody to create detailed expression pattern profiles of their identified cholesterol metabolism targets (scavenger receptor type B class I (SR-BI), ABCA1, and ABC transporter G1 (ABCG1) proteins)1. A research group from the Cornell Medical Center used the ABCA1 antibody to demonstrate that low density lipoprotein receptor (LDLr) deletions found in conditions like atherosclerosis and familial hypercholesterolemia involve cholesterol efflux and ABCA1 macrophage expression2. Bhatnagar et al employed the ABCA1 antibody to help them identify a novel function for the fibroblast growth factor 18 (FGF19) as a hepatic fatty acid synthesis factor in metabolic diseases3.

Zhou’s group used the ABCA1 antibody for immunoblotting and immunohistochemistry in their work on low density lipoprotein receptor related protein (LRP1) and its role as a physiological integrator in lipid metabolism, proliferation, and vascular wall integrity4. ABCA1 antibody flow cytometry studies were done in a Brazilian laboratory, where they examined the role of oxidative stress and ABCA1 in diabetes mellitus5. Their data identifies the cholesterol-mediated signaling mechanism that appears to be responsible for stress-induced atherosclerosis.
