Immunoprecipitation (IP) is a common research technique used to precipitate a specific protein (or protein complex) of interest out of a heterogeneous lysate. The concept of precipitating proteins out of solution is often called a "pull-down". There are a variety of methods to achieve immunoprecipitation, the most common of which uses antibodies bound to magnetic or agarose beads. This process can be used to isolate and concentrate a particular protein or complex from a sample containing many thousands of different proteins, and is often used to study protein-protein interactions through Co-IP experiments.

Rictor silencing was induced by cell transfection with rictor siRNA and then, cells were stimulated with IGF-1 for 24 hours in presence or absence of E2. mTORC2 downregulation was confirmed by abolished Akt-pS473 and resulted in decreased SERCA2A protein expression. ![Western Blot: Rictor Antibody [NB100-612] - mTOR complex protein expressions & rictor phosphorylation at Ser1235. Cardiomyocytes were pretreated for 30 min with 20 nM rapamycin, then stimulated with 10 nM IGF-1 in presence/absence of 10 nM E2 for 24 h. Western blots for GSK3beta-pS9, GSK3beta, rictor pS1235 and rictor. At least 3 independent experiments indicate that rictor phosphorylation at S1235 by GSK-3beta downregulates mTORC2 activity. IGF-1 induced strong phosphorylation of GSK-3beta at Ser9 in the absence and presence of E2. Rapamycin pretreatment reduced phosphorylation in E2 co-treated cardiomyocytes, indicating increased activity of GSK-3beta, which was not associated with increased phosphorylation of rictor at S1235. ![Western Blot: CD63 Antibody (MEM-259) - BSA Free [NB100-77913] - Cytokine & checkpoint molecule arrays from plasma exosomes showed a decreased concentration of IFN-gamma , IL-10, IL-13, B7-1, B7-2, & ICOSL in GBM patients in comparison to normal donors. (A) Quantification of IFN-gamma , IL-10, & IL-13 in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group, *P < 0.05). (B) Quantification of B7-1, B7-2, & ICOSL in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group). (C) Quantification of the immunosuppressive checkpoint & T cell costimulatory homolog protein PD-L1 in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group). Interestingly, PD-L1 is found at similar levels in both normal donor & glioma patient plasma exosomes & these findings were confirmed by western blot. Western Blot analysis also shows that exosomal markers Flotillin-1 & CD63 are found universally in plasma exosomes from normal donors (n = 3), glioblastoma (grade 4) patients (n = 4), & grade 3 glioma patients (n = 3), though CD63 expression may be slightly reduced in glioma patients. ![Immunocytochemistry/Immunofluorescence: LOX Antibody [NBP2-24877] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-LOX (NBP2-24877) at a 1:200 dilution overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue).
