Background

AKR1B10 (aldo-keto reductase 1B10) is a monomeric enzyme that reduces aliphatic and aromatic aldehydes as well as certain hexoses thereby playing a critical role in detoxification of dietary/cellular alpha or beta-unsaturated carbonyls and lipid peroxides (LPOs) ultimately leading to host cells protection against carbonyl lesions. AKR1B10 is active against polycyclic aromatic hydrocarbon (PAHs) which are potent procarcinogens and against all-trans-retinal, the signaling molecule retinoic acid's precursor which regulates cellular proliferation and differentiation. AKR1B10 is a cytosolic protein, but gets secreted through lysosome-mediated mechanisms and in normal tissues, it is expressed in colon, small intestine, adrenal gland etc., whereas its expression gets upregulated in lung bronchial epithelium of cigarette smokers further augmenting the risk of PAHs mediated initiation of carcinogenesis. Moreover, AKR1B10 is overexpressed in certain cancers such as liver, breast and lung cancer, and because of its reducing potential for C13 ketonic groups of daunorubicin and idarubicin, it may provide chemoresistance for cancer cells to these drugs. Moreover, upregulated AKR1B10 in malignant cells blocks ubiquitin-dependent degradation of ACCA (acetyl-CoA carboxylase-alpha) thereby promoting fatty acid/lipid synthesis which is essential to cancer cell growth and proliferation.
