BCL2 was detected in immersion fixed paraffin-embedded sections of human Non-Hodgkin’s Lymphoma using Mouse Anti-Human BCL2, Monoclonal Antibody (Catalog #NBP2-34443) at a 1:500 dilution at 37 ° Celsius for 4 minutes. Before incubation with the primary antibody, tissue underwent an all-in-one dewaxing and antigen retrieval preprocessing using PreTreatment Module (PT Module) and Dewax and HIER Buffer H (pH 9; Epredia Catalog # TA-999-DHBH). TTissue was stained using the Alexa Fluor™ 647 Goat anti-Mouse IgG Secondary Antibody at 1:200 at 37 ° Celsius for 2 minutes. (Yellow; Lunaphore Catalog # DR647MS) and counterstained with DAPI (blue; Lunaphore Catalog # DR100). Specific staining was localized to the membrane. Protocol available in COMET™ Panel Builder.

HIF-1 alpha protein and HIF-1 alpha target genes are elevated in SCs following retrovirus administration. Expression of mRNA for VP16, HIF-1 alpha , and beta -actin in transfected SCs was confirmed by PCR in VP16 and VP16-HIF SCs (A). Protein expression of VP16, HIF-1 alpha , HIF-2 alpha , VEGF, enolase, and protein loading controls (GFP or beta -actin) in transfected SCs was assessed by western blotting (B). Protein expression was normalized to the loading control. Immunohistochemistry-Paraffin: Bcl-2 Antibody (JF104-8) [NBP2-67182] - Analysis of paraffin-embedded mouse kidney tissue using anti-Bcl-2 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody 1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Flow Cytometry: Bcl-2 Antibody (JF104-8) [NBP2-67182] - Analysis of Jurkat cells with Bcl-2 antibody at 1/50 dilution (red) compared with an unlabelled control (cells without incubation with primary antibody; black). Alexa Fluor 488-conjugated goat anti rabbit IgG was used as the secondary antibody.

Western Blot: Bcl-2 Antibody (JF104-8) [NBP2-67182] - Western blot analysis of Bcl-2 on different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1:5,000 dilution was used for 1 hour at room temperature. Positive control: Lane 1: Hela cell lysate Lane 2: Jurkat cell lysate Lane 3: MCF-7 cell lysate

Rabbit anti-Osteopontin was used at a 1:100-1:300 dilution to detect osteopontin by immunohistochemistry. Osteopontin is a normal component of elastic fibers in the breast (shown heavily stained in this section of human breast tumor cells). There is also weak staining of the extracellular matrix. Osteopontin is not expressed in breast tumor cells, and there is no staining of the breast cells in this section. No antigen retrieval is required.
