![Flow Cytometry: Collagen VI alpha 1 Antibody [NB120-6588] - Effect of PTX3 on morphology, collagen production, & receptor expression. After 5 days incubation with 1 ug/ml PTX3, PBMCs were air-dried, fixed, & stained with anti-collagen-I or control antibodies. (Positive staining - red staining; nuclei - blue). Bar is 50 um. Following ICC staining, at least 100 elongated cells with oval nuclei were examined from at least 10 randomly selected fields, & the % of positive cells is expressed as mean +/- SEM (n = 3-5 donors). ![Immunocytochemistry/Immunofluorescence: 58K Golgi Protein Antibody (58K-9) [NB600-412] - HeLa cells, cultured on cover slips, were fixed with 10% formalin for 10 minutes and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were then incubated with 1:200 dilution of anti-58K Golgi Protein antibody (clone 58K-9) for overnight at 4C and detected with an anti-mouse Dylight 488 (Green) secondary at a 1:500 dilution. Alpha tubulin (DM1A) [NB100-690] was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at 1:500 dilution. Nuclei were counterstained with DAPI solution (Blue) [NBP2-31156]. Cells were imaged using a 40X objective. The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Plasma membrane staining was observed. ![Immunohistochemistry-Paraffin: AKT1 [p Ser473] Antibody (104A282) [NB100-56749] - IHC analysis of an FFPE human breast carcinoma tissue section using 1:250 dilution of phospho Ser473 AKT1 antibody (clone 104A282) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10 mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of phosphor-AKT (Ser-473) in the cancer cells as well as the stromal cells.
