Background

RBCk1, also known as HOIL-1L, acts as an E3 ubiquitin-protein ligase, or as part of an E3 complex that contains a zf-RanBP (Zn-finger in Ran binding protein) and a zf-C3HC4 (RING finger) domain. RBCk1 catalyzes the ubiquitination and degradation of IRF3. Both RBCk1 and IRF3 are activated by viral infections and IRF3 is a prerequisite trigger for induction of type I Interferons (1). RBCk1 also negatively regulates TNF and IL1 induced NF-fUB activation (2). Thus it appears RBCk1 and IRF3 interaction represents a controlling antiviral regulatory channel but also that RBCk1 and ubiquitination pathways (3, 4) also are related to NF-kB and its role as an anti-viral response pathway.

A specific band was detected for HOIP/RNF31 at approximately 144 kDa (as indicated) using 5 µg/mL of Mouse Anti-Human HOIP/RNF31 Monoclonal Antibody (Catalog # MAB8039). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.
Western blot shows lysates of Jurkat human acute T cell leukemia cell line and MCF‑7 human breast cancer cell line. PVDF membrane was probed with 0.5 µg/mL of Mouse Anti-Human HOIP/RNF31 Monoclonal Antibody (Catalog # MAB8039) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for HOIP/RNF31 at approximately 120 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
HOIP/RNF31 was detected in immersion fixed MCF‑7 human breast cancer cell line, untreated (upper panel) or treated with Proteasome Inhibitor I (Tocris Bioscience, Catalog # 4045, lower panel), using Mouse Anti-Human HOIP/RNF31 Monoclonal Antibody (Catalog # MAB8039) at 25 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. In vivo assay: IKK gamma Inhibitor Peptide Set [NBP2-26504] - NBD peptide blocks constitutive NF-kB as shown by EMSA. U266 cells were treated with 100 uM of control or NBD peptide for different time periods. Nuclear extracts were isolated and checked for NF-kB-DNA binding activity.
In vivo assay: IKK gamma Inhibitor Peptide Set [NBP2-26504] - NBD peptide blocks constitutive NF-kB activation in human multiple myeloma cells. U266 cells were treated with 100 uM of control (A & B) or NBD peptide (C & D) for 12 hr, cytospun, plated on glass slides, air dried for 1 hr at room temperature and fixed with cold acetone. Slides were blocked with 5% normal goat serum for 1 hr and then incubated with rabbit polyclonal anti-human p65 antibody (A & C) followed by Ig-Alexa 594 second step. In control peptide treated cells, p65 translocates to nucleus (A), whereas NBD peptide prevents translocation of p65 into the nucleus (C). B & D: Nuclear staining with DNA binding dye.
IKK gamma Inhibitor Peptide Set [NBP2-26504] - The IKK complex consists of IKKalpha + IKKbeta+ NEMO. NEMO (IKKgamma/NF-kB Essential MOdulator) is a scaffold protein required for the kinase activity of IKKalpha + IKKbeta. NBD (NEMO Binding Domain) inhibits formation of the IKK complex (IKKalpha + IKKbeta+ NEMO) by binding to NEMO and blocking IKK complex formation. In this regard, NBD is considered to be an 'IKKalpha and IKKBeta decoy' Blocking IKK complex formation suppresses downstream events in the NF-kB signaling pathway that rely on the IKK complex including phosphorylation of IkBalpha, and subsequence IkBalpha ubiquination, degradation and release of NF-kB. Hence, NF-kB is retained in the cytoplasm in its inactive state.

Immunohistochemistry-Paraffin: IRP2 Antibody [NB100-1798] - IHC analysis of a formalin fixed paraffin-embedded (FFPE) human placenta using 1:300 conc. of IRP2 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 30 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 9.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 15 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Cytoplasmic staining was observed in villi of endothelial cells.
