RB1 Antibody (172C1094) [Alexa Fluor® 700] - (cleaved) Summary

• Immunogen: A synthetic peptide corresponding to a region of the caspase-3-induced cleavage site of human Rb was used as immunogen.
• Specificity: The antibody recognizes cleaved Rb (DRb-p70). The antibody does not recognize full-length Rb. In camptothecin treated HL60, an ~68 kDa band is observed. DRb-p70 exists in vivo as an N-terminally truncated form of Rb (Liu et al.). DRb-p70 is thought to be the product of alternative translation and is expressed in primary myeloid cells in fetal liver, bone marrow and spleen. DRb-p70 is also specifically expressed in tumor-associated macrophages (TAMs). TAMs are an important targets of cancer therapy. DRb-p70 is a marker of TAMs, and maybe useful for identifying tumors with macrophage infiltration (Liu et al, 2005).

Background

Rb is a tumor suppressor nuclear phosphoprotein capable of binding to DNA. It is phosphorylated on serine and threonine, but not on tyrosine residues. It forms a complex with SV40 large T antigen, adenovirus E1A, and human papilloma virus-16 E. Rb protein may act by regulating transcription and loss of ts function leads to uncontrolled cell growth. Aberrations in the RB gene have been implicated in cancers of breast, colon, prostate, kidney, nasopharynx, and leukemia.
