Background

p70 S6 kinase (p70S6K, p85S6K) or ribosomal protein S6 kinase has 2 isoforms both of which are monomers. The first isoform, p70, has a predicted molecular mass of 56 kDa and an apparent molecular mass (SDS-PAGE) of 70 kDa. The second isoform, p85 which contains 23 extra residues at the N-terminus, is generated by alternative splicing and has a predicted molecular mass of 59 kDa and an apparent molecular mass of 85 kDa. Serine phosphorylation of the S6 kinase is required for its activation, and this is accompanied by mobility shift in the migration of this 65 kDa kinase on SDA polyacrylamide gels to 70 kDa. Phosphorylation of the S6 protein has been linked with increased rates of initiation of protein synthesis with certain mRNA transcripts.
