Background

The oxytocin receptor belongs to the vasopressin/oxytocin family of GPCRs because it binds similar cyclic nonapeptide hormones. The oxytocin receptor functions primarily during labor and lactation. There is a dramatic increase in the number of oxytocin receptors just before the onset of labor causing the uterine muscles to become exceedingly sensitive to oxytocin. The activated oxytocin receptor causes an inward membrane current that results in smooth muscle contraction during labor and milk ejection during lactation. This receptor also plays a role in natriuresis, kaliuresis and may have an antistress effect. Oxytocin receptor has been reported in breast, central nervous system, heart, ovary, placenta, prostate, salivary gland, testis, uterus, and vessel. ESTs have been isolated from connective tissue, embryo, heart, lung, and melanocyte/uterus/fetal heart, skin, and uterus libraries.

1. I am in need of an antibody that recognizes oxytocin receptors in the rat brain. I will be using this on PFA-fixed free-floating sections (rat). Does the NLS244 work in rat or do you have other antibodies for oxytocin receptor that would work better? In addition, I am looking for a POMC antibody (also to react with rat). Similarly, to work in IHC on fixed free-floating sections. Do Novus have an POMC antibody that will work in rat tissue?
   • NLS244 has only been tested in human tissues. Although the rat and human proteins share a high degree of homology and we expect that there would be cross-reactivity, we cannot guarantee this. We do not currently have an oxytocin receptor antibody that is validated to work on rat samples. For POMC, we have one antibody that we know works in rat samples, NB300-372, but it has never been tested in IHC. In either case, you will qualify for the Innovator's Reward Program should you choose to test an antibody in a species or application that is not listed on the data sheet.
