Western Blot: Mind Bomb 1/MIB1 Antibody (JG69-34) [NBP2-76849] - Analysis of Mind Bomb/MIB1 on SH-SY5Y cell lysates with Rabbit anti-Mind Bomb/MIB1 antibody at 1/500 dilution. Lysates/proteins at 10 ug/Lane. Predicted band size: 110 kDa Observed band size: 110 kDa Exposure time: 2 minutes; 6% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/Immunofluorescence: Mind Bomb 1/MIB1 Antibody (JG69-34) [NBP2-76849] - ICC staining Mib1 in SiHa cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Immunohistochemistry-Paraffin: Mind Bomb 1/MIB1 Antibody (JG69-34) [NBP2-76849] - Analysis of paraffin-embedded human Testis tissue with Rabbit anti-Mind Bomb 1/MIB1 antibody washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

E3 ubiquitin ligase protein mediates ubiquitination of Delta receptors, which act as ligands of Notch proteins. It positively regulates the Delta-mediated Notch signaling by ubiquitinating the intracellular domain of Delta, leading to endocytosis of Delta receptors. It probably mediates ubiquitination and subsequent proteasomal degradation of DAPK1, thereby antagonizing anti-apoptotic effects of DAPK1 to promote TNF-induced apoptosis.
