Background
Leucine-rich repeat flightless-interacting protein 2 (LRRFIP2) belongs to the LRRFIP family. LRRFIP2 has multifaceted functions in innate immunity, apoptosis and development. LRRFIP2 can bind to MyD88 and positively regulate TLR activity as measured by NF-kB activity and cytokine production in macrophages. LPS stimulation also increases LRRFIP2 expression in macrophages. By interacting with Dvl, LRRFIP2 increases the cellular levels of Beta -catenin and activate Beta -catenin/LEF/TCF-dependent transcriptional activity. A dominant negative form of LRRFIP2 suppresses ectopic Wnt signaling in Xenopus embryos and partially inhibits endogenous dorsal axis formation. LRRFIP2 also interacts with Caspase-11 through Flightless-I protein and may be involved in the regulation of proinflammatory caspases by selectively modulating intracellular localization and caspase activity.
