Lgr6 was detected in immersion fixed HEK293 human embryonic kidney cell line transfected with Lgr6 using Mouse Anti-Human Lgr6 Monoclonal Antibody (Catalog # MAB84581) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. In a functional flow cytometry test, biotinylated recombinant human R-Spondin 4 (100 ng/mL) binds to HEK293 human embryonic kidney cell line transfected with human Lgr6 (dark orange filled histogram). Binding is completely blocked (light orange filled histogram) by 1 μg/mL of Mouse Anti-Human Lgr6 Monoclonal Antibody (Catalog # MAB84581). Mouse IgG2A Isotype Control (Catalog # MAB003) at 1 μg/mL was used as a control (blue line). Cells were stained with Streptavidin-APC (Catalog # F0050).

Background

Lgr6 is a glycoprotein hormone receptor phosphorylated upon DNA damage, probably by ATM or ATR. It is a member of the leucine-rich repeat-containing subgroup of the G protein-coupled 7-transmembrane protein superfamily. Lgr6 is a high affinity receptor for R-Spondins 1-3 and potentially functions as a tumor suppressor despite its positive effect on Wnt/ beta -Catenin signaling.
