Background

p29 ING4 is a tumor suppressor protein similar to ING1 that may inhibit tumor progression by modulating the transcriptional output of signaling pathways which regulate cell proliferation. p29 ING4 has been shown to suppress brain tumor angio-genesis through transcriptional repression of RelA/ NFKB3 target genes when complexed with RelA. p29 ING4 may also specifically suppress loss of contact inhibition elicited by activated oncogenes such as MYC. p29 ING4 shows a nuclear localization and interacts with EP300, TP53, RelA, inhibits cell growth, and induces apoptosis. This protein contains a PHD-finger, which is a common motif in proteins involved in chromatin remodeling. Multiple alternatively spliced transcript variants have been observed. The accession number listed below is for variant (1) that encodes the longest isoform.

Western Blot: Bcl-2 Antibody [NB100-56098] - EA reduced MPP+-induced dopaminergic neuronal apoptosis by increasing BDNF (brain-derived neurotrophic factor) expression and further Akt phosphorylation in the rat substantia nigra. Eight days after MPP+ administration, our Western blot results (MAB7566) show that MPP+ treatment reduced tyrosine hydroxylase and Bcl-2 expression in the ipsilateral side of the rat substantia nigra (SN), but not in the contralateral side. EA stimulation (50 Hz) enhanced mature BDNF, tyrosine hydroxylase, and Bcl-2 expression in the MPP+-treated ipsilateral side. Western blot shows lysates of A549 human lung carcinoma cell line . PVDF membrane was probed with 1 µg/mL of Goat Anti-Human ING1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5758) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). Overnight incubation at 4C followed by washes & reaction with a 1:20,000 IRDye(TM)800 conjugated Rb-a-Goat IgG [H&L] MXHu for 45 min at room temperature. IRDye(TM)800 fluorescence image captured using the Odyssey(R) Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (35503, Thermo Fisher).
Immunocytochemistry/Immunofluorescence: KAT3B/p300 Antibody (RW105) [NB100-616] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-KAT3B/p300 Antibody (RW105) NB100-616 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.
