Applications/Dilutions
Dilutions
ELISA 1:10000 - 1:50000
Immunohistochemistry 1:500 - 1:2500
Immunohistochemistry-Paraffin
Western Blot 1:1000 - 1:10000
Application Notes
This secondary antibody has been assayed against 1.0 ug of Swine IgM in a standard capture ELISA using ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:2,000 to 1:8,000 of the reconstitution concentration is suggested for this product. NOTE: Do NOT add Sodium Azide (it inhibits HRP irreversibly).
Store at -20C in the dark. Avoid freeze-thaw cycles.
Buffer
0.02 M Potassium Phosphate, 0.15 M Sodium Chloride, pH 7.2, 10 mg/mL Bovine Serum Albumin (BSA) - Immunoglobulin and Protease free
Preservative
0.01% Gentamicin Sulfate
Purity
Multi-step
Notes
This product is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by cross adsorption and extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Peroxidase, anti-Rabbit Serum, Swine IgM and Swine Serum
Store vial at 4C prior to restoration. For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

FAQs for IgM Antibody (NBP1-73908). (Showing 1 - 1 of 1 FAQ).
Our customer would like to know if NB718 would react to other porcine serum proteins, such as IgG. If there is any further information about its specificity, please share with us. How about other porcine serum proteins, such as IgG? The customer would like to know if NB718 would cross react with IgM, IgA and he also would like to know if it is made from the full-length porcine IgM, or just part of IgM. The customer still has few more questions: 1. The purification of NB718 is described as Immunogen affinity purified on the product page. The customer would like to know further about how the purification is proceeded.2. Based on its datasheet, this antibody can be used for IHC and ELISA. May we have the QC results of NB718 for the customer's reference? 3. Concerning specificity, what the difference is between NB718 and NBP1-73908?
To answer your question, NB718 would react specifically with pig IgM but it may cross react with IgM from other species as well. So NB718 is specific to IgM and would not react with IgG and it was made of heavy chain IgM. With regards to the question No.1, NB718 was isolated by affinity chromatography using antigen coupled to agarose beads and conjugated to horseradish peroxidase (HRP) but in order to provide you with more detailed information on purification process and QC data, I have to contact our lab and I will be in touch with you as soon as I have the answer. NBP1-73908 is an anti-SWINE IgM (mu) antibody that has been generated in rabbit. As far as the specificity is concerned, both antibodies are the same except the host they were generated in.
