HLA DRB1 Antibody (HLA-DRB/1067) - IHC-Prediluted Summary

• Immunogen: Activated human peripheral blood mononuclear cells

• Specificity: This MAb reacts with the beta-chain of HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. Its epitope is different from that of MAb L243. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36kDa alpha (heavy) chain and a 28kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4+ T cells. It is an excellent histiocytic marker in paraffin sections producing intense cytoplasmic staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts. Loss of HLA-DR expression is related to tumor microenvironment and predicts adverse outcome in diffuse large B-cell lymphoma.

• Application Notes: Hu-chromosome location: 6p21.3 Molecular weight of antigen: 28kDa (beta chain) Immunohistochemistry-Paraffin 0.25 - 0.5 ug/ml for 30 minutes at RT; Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10mM Citrate Buffer, pH 6.0, for 10 - 20 min followed by cooling at RT for 20 minutes. The 7mL size is a pre-diluted size and no additional dilutions are required before using this item for the intended application.

1. We are interested in custom labeling our antibodies to be able to integrate new markers into already existing multicolor FACS panels. We would be glad to receive pricing and general order information for anti-HLA-DRB1 and anti-HLA-DRB5. To estimate expenses for future antibody labeling orders it would be helpful to know on what basis (amount, type, etc.) the price is going to be calculated. Right now the antibodies are still in mouse ascites. Would it be possible to integrate the elution of the antibodies from the ascites before labeling as part of the service agreement?
   • While we do not generally do this kind of custom purification unless it is for very large amounts of antibody, we do have a number of kits available that may be of use to you. Since an antibody that is still in ascites can not be labeled, it will need to be purified. A list of our kits available for this can be found at the following link: AbSelect.
