Background
GPR119 (G-protein coupled receptor 119) is a class A (rhodopsin-like) 7-transmembrane GPCR with little homology to other receptors, and human GPR119 receptor has 335 amino acids which is highly conserved in mouse and rat. GPR119 is expressed in human insulin-secreting pancreatic islets and incretin (GIP and GLP-1) secreting K- and L-cells of gastrointestinal tract. GPR119 has been suggested to function as a glucose-dependent insulinotropic receptor and its activity is mediated by G proteins which activate adenylate cyclase. Lysophosphatidylcholine (LPC), oleoylethanolamide (OEA), N-acylated ethanolamine phospholipids (NAPEs) and N-oleoyldopamine (OLDA), and 5-hydroxyeicosapentaenoic acid (5-HEPE) have all been proposed as GPR119's endogenous ligands. Moreover, GPR119 agonists have been shown to elicit nutrient-independent incretin and gastrointestinal hormone release as well as nutrient-stimulated insulin secretion, thereby lowering plasma glucose levels during an oral glucose tolerance test. In rodent models, GPR119 mediates improved glucose tolerance and prevents weight gain. GPR119 agonism increases the release of incretin hormones and insulin from enteroendocrine and pancreatic beta cells which may underlie these beneficial effects. GPR119 agonists play a role in reducing food intake and promoting weight loss, possibly through the release of GLP-1, which is known to delay gastric emptying.
