Immunohistochemistry-Paraffin: Glucagon Antibody (JF0960) [NBP2-66869] - Immunohistochemical analysis of paraffin-embedded rat pancreas tissue using anti-Glucagon antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (1/200) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

The protein encoded by the Glucagon gene is actually a preproprotein that is cleaved into four distinct mature peptides. One ofthese, glucagon, is a pancreatic hormone that counteracts the glucose-lowering action of insulin by stimulatingglycogenolysis and gluconeogenesis. Glucagon is a ligand for a specific G-protein linked receptor whose signallingpathway controls cell proliferation. Two of the other peptides are secreted from gut endocrine cells and promotenutrient absorption through distinct mechanisms. Finally, the fourth peptide is similar to glicentin, an activeenteroglucagon. (provided by RefSeq)
