Immunoprecipitation: FAT4 Antibody [NBP2-78795] - Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-FAT4 antibody NBP2-78795 used for IP at 15 ul per reaction. FAT4 was also immunoprecipitated by rabbit anti-FAT4 antibody from Company B. For blotting immunoprecipitated FAT4, NBP2-78795 was used at 1:1000. Detection: Chemiluminescence with an exposure time of 75 seconds.

Background

FAT4 (FAT tumor suppressor homolog 4, also know as Fat-J, mFat4, and CDHF14) is a member of the Cadherin superfamily. Members of the Cadherin superfamily positively regulate planar cell polarity (PCP) in drosophila wings and play an important role in cell to cell adhesion interactions. FAT4 and other Cadherin family members have been used in systems medicine; especially in the fields of oncology and neurology (PMID: 16865240). FAT4 is widely expressed in most tissues, especially fetal and infant brain. It is also expressed in brain tumors and colorectal cancer. Furthermore, human breast tumor cell lines and primary tumors show a loss of FAT4 expression. Promoter methylation of a FAT4 allele can inactivate the expression of the FAT4 gene in deleted regions of the Hippo signaling pathway, the pathway that controls organ size (PMID: 19048595).
