ER beta/NR3A2 Antibody

Western Blot: ER beta/NR3A2 Antibody [NB300-747] - Recombinant full-length human ER alpha & ER beta proteins, & 10ul of PageRuler Prestained Protein Ladder loaded onto a Novex 4-20% Tris-Glycine polyacrylamide gel. Transferred to PVDF membrane using G2 Blotter, & blocked with 5% milk in TBST for 1 hr at RT. ER beta at ~54kD (left) using ER beta pAb at 1:1000 in blocking buffer overnight at 4C on rocking platform, then HRP- goat anti-rabbit IgG secondary antibody at 1:40000. Chemiluminescent detection performed using SuperSignal West Pico. The blot stripped with Restore Plus Western blot stripping buffer for 15 min at room temp, re-blocked with 5% milk in TBST for 1 hr at room temp, & re-probed with an ER alpha pAb at a dilution of 1:2000 overnight at 4C, followed by an HRP-goat anti-rabbit IgG secondary antibody at a dilution of 1:40,000. Chemiluminescent detection was performed using SuperSignal West Pico. The presence of faint ER-beta b&s on the blot probed.

Background

Steroid receptors are ligand-dependent intracellular proteins that stimulate transcription of specific genes by binding to specific DNA sequences following activation by the appropriate hormone.
