Western blot shows lysates of mouse skin tissue and mouse embryo (15 d.p.c.) tissue. PVDF membrane was probed with 1 µg/mL of Rat Anti-Mouse Desmocollin‑3 Monoclonal Antibody (Catalog # MAB7265) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). Specific bands were detected for Desmocollin‑3 at approximately 95-110 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Desmocollin‑3 was detected in perfusion fixed frozen sections of mouse embryo (15 d.p.c.) using Rat Anti-Mouse Desmocollin‑3 Monoclonal Antibody (Catalog # MAB7265) at 25 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Rat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS017) and counterstained with hematoxylin (blue). Western Blot: Desmoglein-3 Antibody (5H10) [NBP1-78984] - Colocalisation of Dsg3 with E-cadherin-catenin complex. Co-IPs demonstrated that both endogenous and exogenous Dsg3 interacted with the E-cadherin-catenin complex in A431 stable lines (the empty vector control (V) and hDsg3.myc (D3)) as well as in HaCaTs. When pulled down with antibodies against the myc tag or with a mouse Dsg3 Ab 5H10 and blotted for E-cadherin and beta-catenin, both proteins were detected in D3 and, to a lesser extent, in V. Conversely, when pulled down with E-cadherin Ab HECD-1 and blotted for Dsg3 or the myc tag and Pg, these proteins were detected in both V and D3 cells. Note that this result also was confirmed in HaCaTs (HaC) where Dsg3 was expressed endogenously (far right blot). Flow Cytometry: EGFR Antibody (DH8.3) - EGFRvIII [NBP2-50599] - An intracellular stain was performed on A431 cells with EGFR [DH8.3] Antibody NBP2-50599R (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to DyLight 550.
Flow Cytometry: EGFR Antibody (DH8.3) - EGFRvIII [NBP2-50599] - An intracellular stain was performed on A431 cells with EGFR [DH8.3] Antibody NBP2-50599AF700 (blue) and a matched isotype control (orange). Western Blot: beta-Catenin Antibody (12F7) [NBP1-54467] - Total protein from human HepG2 and HeLa cells, rat PC12 cells and mouse 3T3 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 1.0 ug/ml anti-Beta catenin in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Western Blot: beta-Catenin Antibody (12F7) [NBP1-54467] - Activity of Wnt/beta-catenin pathway in the embryonic chick lung. Western blot analysis of active and total beta-catenin in stage b1, b2 and b3 lungs, and stage 24 limb (as positive control). Control loading was performed using beta-tubulin (55 kDa). Total and active beta-catenin correspond to 92 kDa.
