Specificity
Recognizes a single chain glycoprotein of 105/120kDa, identified as CD30/Ki-1. Its epitope is located between aa112-412. CD30 is synthesized as a 90kDa precursor, which is processed in the Golgi complex into a membrane-bound phosphorylated mature 105/120kDa glycoprotein. In Hodgkins disease, CD30/Ki-1 antigen is expressed by mononuclear-Hodgkin and multinucleated Reed-Sternberg cells. It is also expressed by the tumor cells of a majority of anaplastic large cell lymphomas as well as by a varying proportion of activated T and B cells. This monoclonal antibody distinguishes large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and lymphomas derived from resting and precursor lymphoid cells or from anaplastic carcinomas. About one third of the Ki-1 positive lymphomas lack the leukocyte common antigen (CD45).

Immunohistochemistry (Formalin-fixed): 1:50-1:100 for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 1mM EDTA, pH 7.5-8.5, for 45 min at 95C followed by cooling at RT for 20 minutes
The 7mL size is a pre-diluted size and no additional dilutions are required before using this item for the intended application.

Background

CD30 is a type I transmembrane protein and a member of the tumor necrosis factor receptor superfamily of proteins. (1,2) Murine CD30 is expressed predominantly in the thymus, and it is inducible in mouse splenocytes stimulated with pokeweed mitogen or concanavalin A. (1) In anti-CD3epsilon-activated spleen cells, CD30 is expressed primarily on the surface of CD8+ T cells with peak expression on days 4 and 5. Stimulation of CD30+ CTL lines with plate-bound anti-CD30 directly signals IL-5 but not IFN; production. (1) While these studies demonstrate that CD30 directs cytokine secretion and suggest that CD30 may play a pivotal role in the pattern of cytokine production by T cells, the precise roles of CD30 and its ligand (CD153) in T-cell development have not been clearly defined.
