Staining seen in cytoplasm, ER and membrane. Fixation in 95% ethanol/5% acetic acid for 2-3 hours prior to paraffin embedding is recommended. Specimens which have not been fixed in acetic acid/alcohol require pre-treatment with citrate buffer: for example, unmask of the target epitope by boiling the tissue sections in 10mM citrate buffer, pH 6.0, for 10 min followed by cooling at room temp for 20 min.

Western Blot: p97/VCP Antibody [NB100-1558] - Zymographic assay of gelatinase activity. Medium supernatants of 20ml culture medium (medium control) as well as 20 ml conditioned medium from 107 U937 cells, pMTH1-U937 cells and asCD11b-U937 cells in the absence or presence of 5nM TPA for 72 h, respectively, were 18-fold concentrated and subjected to SDS-PAGE containing 2 mg/ml of gelatine. Following incubation with MMP enzyme buffer, the gels were stained with 0.4% Coomassie blue and afterwards destained again to visualize the appearance of gelatinase activity by light bands against the dark background. The molecular weight markers on both sides of the gels indicate the size of the MMPs exhibiting gelatinase activities. Immunocytochemistry/Immunofluorescence: beta-Synuclein Antibody [NBP3-05529] - Immunofluorescent analysis of mouse hippocampus section stained with beta-Synuclein Antibody at a at a dilution of of 1:500 in green, and costained with rabbit polyclonal antibody to C-terminal peptide of rat NF-L at a at a dilution of of 1:5,000, in red. The blue is Hoechst staining of nuclear DNA. Following transcardial perfusion of mouse with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45uM, and free-floating sections were stained with above antibodies. The beta-Synuclein Antibody detects protein concentrated in synaptic regions, while the NF-L antibody labels dendrites and axons of neuronal cells.

Knockdown Validated: PINK1 Antibody [BC100-494] - HEK293 cells were co-transfected with PINK1 siRNA (#1 or #2) or scrambled siRNA (scrambled) and untagged wild-type (WT) or Ser65Ala (S65A) mutant Parkin as indicated using TransFectin reagent (Bio-Rad). 48 hrs post-transfection, cells were treated with/without 10 uM CCCP for 3 h. 0.25 mg of 1% Triton whole-cell lysate were subjected to immunoprecipitation with GST-Parkin antibody (S966C) covalently coupled to protein G Sepharose and then immunoblotted with anti-phospho-Ser65 antibody in the presence of dephosphorylated peptide. 5% of the IP was immunoblotted with total anti-Parkin antibody. 0.25 mg of whole-cell lysates were immunoprecipitated with anti-PINK1 antibody (S085D) and immunoblotted with anti-PINK1 antibody.
