Western Blot: 14-3-3 Antibody (SD084-02) [NBP2-67831] - Analysis of 14-3-3 Theta on different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody ( 1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:5,000 dilution was used for 1 hour at room temperature. Positive control: Lane 1: MCF-7 cell lysate Lane 2: Hela cell lysate

Flow Cytometry: 14-3-3 Antibody (SD084-02) [NBP2-67831] - Analysis of SH-SY-5Y cells with 14-3-3 Theta antibody at 1/50 dilution (red) compared with an unlabelled control (cells without incubation with primary antibody; black). Alexa Fluor 488-conjugated goat anti rabbit IgG was used as the secondary antibody.

Background

14-3-3 family of proteins is composed of seven isotypes, beta, gamma, zeta, epsilon, tau, eta, and sigma, that play critical roles in cell signaling events that control cell cycle progression, transcriptional alterations, and apoptosis (1-4). 14-3-3 proteins were the first signaling molecules to be identified as specific phosphoserine/threonine binding proteins (1). 14-3-3 can serve as a direct regulator of its target by altering the function of the protein (3). 14-3-3 protein targets include the signalling intermediates Raf, MEKK, PI-3 kinase and IRS-1, cell cycle proteins CDK2, Wee1, and Cdc25, and apoptosis proteins BAD and ASK-1 (3,4).
