<b>Biological Strategies Validation. </b>CXCR4 neutralization leads to attenuation of the CD44+/CD133+ prostate progenitor population.(A) DU145 cells were plated in 96-well low-attachment plates at 100 cells per well (5 replicates) and the spheres were grown in serum-free, EBM medium with supplements. The antibodies were replenished daily. Cells were imaged with an Acumen eX3 microplate cytometer and spheres were detected using image analysis software. The sphere size was measured by GFP intensity. The spheres were discriminated from cell debris using a Gaussian filter. The spheres included in the analysis are outlined in red and indicated by arrows. Representative data from one of two independent experiments is shown; *- p value<0.05. (B) Flow cytometry analysis revealed attenuation of CD44+/CD133+ population in DU145 cells treated with 10 µg/ml neutralizing anti-CXCR4 (mouse monoclonal IgG, clone 44716, R&D Systems) or 10 µg/ml control antibody (mouse IgG isotype control, Lifespan Bioscience Inc.) for 5 days. Knockdown of CXCR4 abrogates trastuzumab resistance. CXCR4 was silenced by specific shRNA in HCC1419 cells (“Materials and methods” section). Immunoprecipitation: Complement C3 Antibody [NBP1-32080] - Immunoprecipitation of C3 protein from HepG2 whole cell extracts using 5 ug of C3 antibody [C3], C-term.

Human plasma (30 ug) was separated by 7.5% SDS-PAGE, and the membrane was blotted with Complement C3 antibody [C3], C-term (NBP1-32080) diluted at 1:10000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.

Western Blot: CXCR4 Antibody [NB100-74396] - Analysis of CXCR4 protein in human small intestine tissue lysate using 1:500 dilution of rabbit polyclonal CXCR4 antibody (Lot A2). The signal was developed using ECL method and the antibody generated a specific band of CXCR4 at ~40 kDa position.

$Immunocytochemistry/ Immunofluorescence: CXCR4 Antibody [NB100-74396] - Isolation of prostate CTCs from metastatic PCa patients using anti-CD45 immunomagnetic depletion.2.5 ml blood from three metastatic PCa patients (> 50 CTCs/ 2.5 ml blood) was processed via ficoll density centrifugation & the PBMC fraction was collected. Immunomagnetic anti-CD45 depletion was performed on the obtained PBMCs & the remaining cells were washed, cytospunned onto the slides. Slides were stained for PSMA, EpCAM, sLex, & CXCR4 using the protocol as described in Figure S1. MDA, PC3, & KG1 cells were simultaneously stained as a control for the following markers: PSMA= Magenta, EpCAM= Yellow, HECA-452= Green, CXCR4= Red. All prostate CTCs expressed CXCR4, while, sLex expression was variable. The analysis of sLex intensity is shown in Figure 4.
