(A) BMDM were unchallenged (control) or challenged with 500 HU/mL SLO WT, PFO or an equivalent mass of SLO ML for 10 min at 37 °C & centrifuged at 2000 × g for 5 min to yield cell pellet (C). The TLR4/MD-2/CD14/IL-8 Prom/LUCPorter HeLa cells were plated in 96-well white plates at 1 x 10^5 cells/well. After 16 h, cells were preincubated with different amounts of this antibody for 1 h and then stimulated with 10 ng/ml LPS for 6 h. Luciferase activity was then directly analyzed using the one-step luciferase reporter assay reagent. Data Summary: It inhibited LPS-mediated TLR4 activity in the TLR4/MD-2/CD14/IL-8 Prom/LUCPorter cell line. ![Immunocytochemistry/Immunofluorescence: TLR4 Antibody [NBP2-24821] - U2OS cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-TLR4 [NBP2-24821] at a 1:50 dilution overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse DyLight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue).
