Enzyme-linked immunosorbent assay (ELISA) is an analytic biochemical assay that utilizes enzyme-linked antibodies on a microwell platform to quantitate specific proteins within biological samples. Visual detection is then achieved by adding the enzyme’s substrate, which causes a color-changing reaction to occur. There are three different types of ELISA that are most commonly used: indirect ELISA, sandwich ELISA and competitive ELISA. These assays are extremely useful for high-throughput diagnostic screenings drug testing.

![Immunoprecipitation: Dynamin Antibody (3G4B6) [NB110-60491] - Antibody-bead conjugates were prepared by adding 1.0 ug of antibody to 500 ul of PBS with 0,01% triton X-100 in a microcentrifuge tube and 30ul of protein G Sepharose beads. Tubes were rocked O/N at 4 degrees C followed by several washes to remove unbound antibodies. U2OS were collected in HEPES buffer (20 mM HEPES, 100 mM sodium chloride, 1 mM EDTA, 1% Triton X-100, pH 7.4) supplemented with protease inhibitor. Lysates are rocked 30 min at 4 degrees C and spun at 110,000xg for 15 min at 4 degrees C. One ml aliquots at 1 mg/ml of lysate were incubated with an antibody-bead conjugate for ~2 hrs at 4 degrees C. Following centrifugation, the unbound fractions were collected, and beads were subsequently washed three times with 1 ml of HEPES lysis buffer and processed for SDS-PAGE and immunoblot. Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 15 minutes at room temperature. Cells were then blocked with 5% normal goat serum for 15 minutes at room temperature. Cells were probed with a mouse monoclonal antibody recognizing CREB at a dilution of 1:400 for at least 1 hour at room temperature. Cells were washed with PBS and incubated with DyLight 488 goat-anti-mouse secondary antibody at a dilution of 1:400 for 30 minutes at room temperature. ![Immunoprecipitation: CREB Antibody (LB9) [NB100-74393] - Analysis of CREB was performed on untreated 293T cells. Antigen: antbody complex was formed by binding 500ug whole cell lysate with 2ug of mouse monoclonal antibody recognizing CREB overnight on a rocking platform at 4C. The immune-complex was then captured on 50ul Protein A/G Plus Agarose. Captured immune-complexes were then washed extensively and proteins eluted with 5X Reducing Sample Loading Dye. Samples were then resolved on a 4-20% Tris-HCl polyacrylamide gel. Proteins were transferred to PVDF membrane and blocked with 5% Milk/TBS-0.1%Tween for at least 1 hour. Membranes were then probed with a mouse monoclonal antibody recognizing CREB at a dilution of 1:1000 overnight rotating at 4C. Membranes were then washed in TBST and probed with a goat anti-mouse-HRP secondary antibody at a dilution of 1:20,000 for at least one hour.
