![Flow (Intracellular): Complement C3 Antibody (11H9) [NB200-540] - An intracellular stain was performed on RAW 246.7 cells with Complement C3 (11H9-3-2) antibody NB200-540 (blue) and a matched isotype control NBP2-31382 (orange). Cells were either treated with 3uM Monensin for 3 hours to block the secretion of Complement C3 (B) or grown in normal media (A). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. ![Immunocytochemistry/Immunofluorescence: Complement C3 Antibody (JF10-30) [NBP2-66994] -Cells were fixed in 100% precooled methanol for 5 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-C3 antibody at 1/100 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (iFluor™ 488, was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI.Beta tubulin (M1305-2, red) was stained at 1/100 dilution overnight at +4 c.
