The immune-complex was then captured on 50ul Protein A/G Plus Agarose. Captured immune-complexes were then washed extensively and proteins eluted with 5X Reducing Sample Loading Dye. ![Western Blot: BDNF Antibody [NBP1-59304] - Simvastatin treatment increases BDNF expression in primary cortical neurons of AS mice. (A) Primary cultured cortical neurons prepared from wild type & AS embryos were treated with 5 μM simvastatin at DIV14 for 12 h. Neurons were then fixed & processed for double immunofluorescence staining using Ube3a & BDNF antibodies. Representative images were shown. About 12 immunostained neurons in each group were checked for BDNF expression, fluorescence intensity in each cell was quantified & compared. Scale bar, 50 μm. (B) Immunoblot analysis of matured BDNF levels in simvastatin treated primary cortical neurons as described above. ![Chromatin Immunoprecipitation: CREB Antibody [NBP2-22516] - Analysis performed using cross-linked chromatin from colon carcinoma cells treated with serum. IP was performed using a multiplex microplate Matrix ChIP assay of a CREB polyclonal antibody. Chromatin aliquots from cells were used per ChIP pull-down. Quantitative PCR data one in quadruplicate using 1ul of eluted DNA in 2ul SYBR real-time PCR reactions containing primers to amplify -15kb upstream of Egr1 or exon-1 or exon-2 of Egr1.Quantitation of immunoprecipitated chromatin is presented as signal relative to the total amount of input chromatin.
