![Immunohistochemistry-Paraffin: SCP3/SYCP3 Antibody [NB300-232] - IHC analysis of a formalin fixed and paraffin embedded tissue section of Mouse testis using SCP3 antibody at 1:1000 dilution. The signal was detected using HRP-DAB detection method and the nuclei were counterstained with hematoxylin. The antibody generated a specific signal of SCP3 in the spermatogonial cells and the spermatocytes. SYCP3 is a protein essential for synapsis of homologous chromosomes & gamma H2AX localizes to double-strand breaks & XY bodies during meiosis. A total of 200 spreads were counted in testis cell preparations from WT & GCKO mice at P22. (B) Chi square analysis shows GCKO testes contained significantly higher numbers of germ cell spreads at the leptotene & zygotene stages of meiosis I & fewer spreads at pachytene, diplotene & metaphase I stages (P<0.001), suggesting that the loss of Dicer1 lead to disruptions in progression through meiosis I. ![<b>Independent Antibodies Validation. </b>Immunoprecipitation: Cdc20 Antibody [NB100-59828] - Detection of human CDC20 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-CDC20 antibody NB100-59828 (lot NB100-59828-2) used for IP at 6 ug per reaction. CDC20 was also immunoprecipitated by a previous lot of this antibody (lot NB100-59828-1) and rabbit anti-CDC20 antibody NB100-59827. For blotting immunoprecipitated CDC20, NB100-59828 was used at 1 ug/ml.
