Background
Tissue plasminogen activator (tPA) is a serine protease which converts the abundant proenzyme plasminogen to active plasmin. It plays a key role in thrombolysis (1) and several neurobiological processes (2).

I have just purchased one of your antibodies for human tPA (catalog # NBP1-05159). I also noticed this comment on you product sheet: "T-1 reacts with non-reduced tPA but not with reduced tPA in Western blotting. It inhibits the enzymatic activity of tPA. For immunohistochemistry applications T-1 can be used on frozen sections." Does that mean that the antibody will only recognized non-denatured protein? Or it will indeed recognize denatured protein, as long as it is not reduced. If so, I should assume that it will work by simply excluding DTT from the loading buffer. I truly hope this antibody does not work only for native gels.
- This antibody should recognize the denatured, but non-reduced protein. So you are correct, just exclude DTT and you can proceed as normal.
