Background

Somatostatin Receptor type 5 (SSTR5) is one of five 7-transmembrane G-protein-coupled receptors for somatostatins 14 and 28. Within the extracellular portions, human SSTR5 shares 65% and 68% aa identity with mouse and rat SSTR5, respectively. It is expressed in adult pituitary, heart, small intestine, adrenal gland, cerebellum and fetal hypothalamus. It is an inhibitory receptor that transduces the antiproliferative and antisecretive effects of somatostatins.

Immunocytochemistry/Immunofluorescence: Somatostatin R2/SSTR2 Antibody [NB300-157] - U2OS cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton X-100. The cells were incubated with antibody at 1:50 overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at 1:500. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Recombinant Human Growth Hormone protein was serially diluted 2-fold and captured by Mouse Anti-Human Growth Hormone Monoclonal Antibody (Catalog # MAB10671) coated on a Clear Polystyrene Microplate (Catalog # DY990). Goat Anti-Human/Mouse/Rat Growth Hormone Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1067) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998) followed by Substrate Solution (Catalog # DY999) and stopping the enzymatic reaction with Stop Solution (Catalog # DY994).
Recombinant Human Growth Hormone (Catalog # 1067-GH) stimulates proliferation in the Nb2‑11 rat lymphoma cell line in a dose-dependent manner (orange line). Proliferation elicited by Recombinant Human Growth Hormone (0.2 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human/Mouse/Rat Growth Hormone Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1067).
