Background

Rb is a tumor suppressor nuclear phosphoprotein capable of binding to DNA. It is phosphorylated on serine and threonine, but not on tyrosine residues. It forms a complex with SV40 large T antigen, adenovirus E1A, and human papilloma virus-16 E. Rb protein may act by regulating transcription and loss of ts function leads to uncontrolled cell growth. Aberrations in the RB gene have been implicated in cancers of breast, colon, prostate, kidney, nasopharynx, and leukemia.

1. My research group wants to order a Retinoblastoma 1 Antibody for IHC-Paraffin. We already bought 2 antibodies from other companies and they didn't work properly. There was always a cytoplasmatic staining and no nuclear staining. I found two antibodies on your website: Retinoblastoma 1 Antibody (7E4B8) (NBP1-51527) (0.1 ml) and Retinoblastoma 1 Antibody (IF8) (NB100-65771) (0.1 ml). Can you suggest which antibody we should buy? We are using a VENTANA BenchMark ULTRA stainer, FYI. We stain human material.

2. Our customer would like to know if NB120-3077 would recognize both phosphorylated and non-phosphorylated Retinoblastoma 1, or only non-phosphorylated Retinoblastoma 1?
   • Retinoblastoma 1 Antibody (1F8-Rb1), catalog number NB120-3077, will recognize both phosphorylated as well as non-phosphorylated Retinoblastoma 1. Technically speaking, only antibodies raised to specific phospho-site recognize JUST phosphorylated form of a given target. Non-phospho specific antibodies detect total target (phospho + non-phospho).
3. Hi could you by chance tell me what region was used to raise this antibody: Retinoblastoma 1 Antibody (1F8-Rb1) or if it was raised against full length.
