Western Blot: RanBP16 Antibody [NBP2-76384] - Samples: Whole cell lysate (15 ug) from Jurkat, A-549, Hep-G2, mouse TCMK-1, and mouse NIH 3T3 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-RanBP16 antibody NBP2-76384 used for WB at 1:1000. Detection: Chemiluminescence with an exposure time of 30 seconds.

Immunoprecipitation: RanBP16 Antibody [NBP2-76384] - Detection of human RanBP16 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from Jurkat cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-RanBP16 antibody NBP2-76384 used for IP at 100 ul per reaction. RanBP16 was also immunoprecipitated by another rabbit anti-RanBP16 antibody. For blotting immunoprecipitated RanBP16, NBP2-76384 was used at 1:40. Detection: Chemiluminescence with an exposure time of 3 seconds.

Background

RanBP16 mediates the nuclear export of proteins (cargos) with broad substrate specificity. In the nucleus binds cooperatively to its cargo and to the GTPase Ran in its active GTP-bound form. Docking of this trimeric complex to the nuclear pore complex (NPC) is mediated through binding to nucleoporins. Upon transit of a nuclear export complex into the cytoplasm, disassembling of the complex and hydrolysis of Ran-GTP to Ran-GDP (induced by RANBP1 and RANGAP1, respectively) cause release of the cargo from the export receptor. RanBP16 then return to the nuclear compartment and mediate another round of transport. The directionality of nuclear export is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus.
