Background

Raf-1(v-raf-1 murine leukemia viral oncogene homolog 1), with 648-amino acid protein (about 71 kDa), is a MAP kinase kinase kinase (MAP3K) which functions downstream of the Ras family of membrane associated GTPases to which it binds directly. The Raf family of serine/threonine specific kinases is comprised of three members (aRaf, bRaf, and Raf-1).The Ras-Raf-MEK-extracellular signal-regulated kinase (ERK) pathway participates in the control of many fundamental cellular processes including proliferation, survival, and differentiation. The pathway is deregulated in up to 30% of human cancers, often due to mutations in Ras and the bRaf isoform. Raf-1 and bRaf can form heterodimers, and this may be important for cellular transformation. Raf-1 inhibition is also associated with apoptotic cell death.
