Background
GPR54 is a Metastin Receptor. Metastin, also known as KiSS-1, is a metastasis suppressor gene. Mutations in GPR54 have been shown to cause autosomal recessive idiopathic hypogonadotropic hypogonadism in humans and mice, suggesting that GPR54 is a key regulator for the onset of puberty (Seminara et al. 2003). GPR54 has been reported in various human brain regions, placenta, pancreas, and spinal cord, and at lower levels in spleen, peripheral blood leukocytes, testis, lymph node, thymus, small intestine, stomach, lung, and kidney. The receptor is also abundant in tumor tissues. In rat, GPR54 is expressed in liver, intestine, and in the pons, midbrain, thalamus, hypothalamus, hippocampus, amygdala, cortex (including frontal), and striatum. ESTs have been isolated from normal placenta and kidney cancer libraries.

We would like to know the sequence of the peptide against which the GPR54 antibody was generated. Furthermore, it is not clear how the antibody is purified. Is it affinity purified? And are thus removed of all IgGs specific IgGs against the peptide?
The immunizing peptide sequence is proprietary and cannot be disclosed. NLS1926 is affinity purified, which means the immunogen was used to isolate the antibody removing all other Ig's.
For GRP54 antibody # NLS1926, what could one use as POSITIVE CONTROL?
According to the Uniprot listing, levels of GPR54 are highest in pancreas, placenta and spinal cord (UniProt Q969F8). Depending on your method of detection, we have a number of products that you could use as a positive control. Do you know what tissue they used in testing their antibody? The pictures are too small to see what is staining (in the cerebellum and tonsil) and the type of staining. As my study is on placentas, I will obviously need another control! The kisspeptin antibody mentions prostate (AbDSerotec).
The images generated with the GPR54 antibody were from formalin-fixed paraffin-embedded human tonsil and brain (cerebellum) samples. I'm sorry that we have no other images which we can share with you at the current time. The Uniprot listing for GPR54 makes no mention of the levels of this protein in prostate. If your customer is interested, we do sell prostate tissue lysates, from normal and tumour conditions, although we only currently sell one (diseased type) prostate tissue slide, with catalogue number S162W0. We sell two placenta lysates - NB820-60607.html and NB820-59248 - however according to Uniprot, expression levels of GPR54 are higher in early (7-9 weeks) than term placentas, and these are adult samples. We unfortunately do not currently stock any placenta tissue slides.

Would it be possible to know the amino acid sequence of the immunogen used to produce this antibody?
It was raised against a synthetic 17 amino acid peptide which was derived from the second extracellular domain of the human protein (the immunogen lies within amino acids 102-120).
