Background

Interferon induces a variety of effects on target cells including antiviral, antiproliferative, and immunomodulatory activities. Interferon alpha-2b can either inhibit or enhance immune cell activity within the tightly regulated microenvironment of inflammation, depending upon the concentration of the cytokine and the activation stage of the cell. Interferon alpha-2b was expressed in E.coli and purifed by conventional chromatography, after refolding of the isolated inclusion bodies in a renaturation buffer.

  1. Why does interferon stand under 18 KD in SDS-PAGE gel?
    • The molecular weight of 19.4 kDa for NBC1-18467 has been confirmed by MALDI-TOF. The migration and hence exact band position of a target protein in western blot can be influenced by many factors, such as different gel types and buffers. As the observed band is only slightly different from the theoretical molecular weight, I would think that this is the likely explanation. The lab have now replied to me in answer to your enquiry. They say, as I noted, that although size is the major determinant of band migration, other factors can affect it too to a certain extent (they give charge as another example). They also point out that the difference in this case is very small, and slight mis-alignment with the marker could be a cause. Finally, they emphasize that SDS-PAGE, MALDI-TOF and DNA sequence analysis of the protein has confirmed its composition.
