• Specificity: This monoclonal antibody reacts with the HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. It binds a conformational epitope on HLA-DR, which depends on the correct folding of the heterodimer. This monoclonal antibody has been reported to block mixed lymphocyte reactions. The L243 antibody recognizes a different epitope than the LN3 monoclonal antibody, and these antibodies do not cross-block binding to each others respective epitopes. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36kD alpha (heavy) chain and a 28kD beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4+ T cells. It is an excellent histiocytic marker in paraffin sections producing intense staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts.

1. We are interested in custom labeling our antibodies to be able to integrate new markers into already existing multicolor FACS panels. We would be glad to receive pricing and general order information for anti-HLA-DRB1 and anti-HLA-DRB5. To estimate expenses for future antibody labeling orders it would be helpful to know on what basis (amount, type, etc.) the price is going to be calculated. Right now the antibodies are still in mouse ascites. Would it be possible to integrate the elution of the antibodies from the ascites before labeling as part of the service agreement?
   • While we do not generally do this kind of custom purification unless it is for very large amounts of antibody, we do have a number of kits available that may be of use to you. Since an antibody that is still in ascites can not be labeled, it will need to be purified. A list of our kits available for this can be found at the following link: AbSelect.
