Background

The granzyme family of proteins belong to the larger peptidase S1 family. Granzyme A and granzyme B are serine proteases that facilitate apoptotic signaling in cytotoxic T lymphocytes (CTL) and natural killer (NK) cells. Within the granules of activated CTLs, granzyme A and B are processed and converted to their active forms by the lysosomal cysteine protease cathepsin C. Once cleaved, these active proteases target distinct substrates for proteolysis and, thereby, mediate apoptosis through two different pathways. Granzyme H localizes to cytoplasmic granules of cytolytic T lymphocytes and is important for target cell lysis in cell-mediated immune responses. Granzyme K (GMZK), also designated granzyme-3 or NK-tryptase-2 (NK-TRYP-2), contains one peptidase S1 domain. Granzyme K is a serine protease localizing to the granules of natural killer cells and cytotoxic T lymphocytes. It is primarily expressed in thymus, lung, spleen and peripheral blood leukocytes.

Recombinant Human Granzyme&amp;nbsp;A protein was serially diluted 2-fold and captured by Mouse Anti-Human Granzyme&amp;nbsp;A Monoclonal Antibody (Catalog&amp;nbsp;# MAB29052) coated on a Clear Polystyrene Microplate (Catalog&amp;nbsp;# DY990). Goat Anti-Human Granzyme&amp;nbsp;A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2905) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog&amp;nbsp;# DY998) followed by Substrate Solution (Catalog&amp;nbsp;# DY999) and stopping the enzymatic reaction with Stop Solution (Catalog&amp;nbsp;# DY994).
Western blot shows lysates of NK‑92 human natural killer lymphoma cell line. Immunohistochemistry-Paraffin: Caspase-1 Antibody (14F468) [NB100-56565] - Tissue section of human intestine using Caspase-1 antibody (clone 14F468) at 5ug/ml concentration (1:200 dilution). The primary antibody binding to Caspase 1 in cells was detected using HRP conjugated anti-Mouse secondary antibody with DAB reagent, and the sections were further counterstained with hematoxylin for labeling cellular nuclei. This Caspase 1 antibody generated a diffused but specific cytoplasmic staining in columnar epithelia cells of villi, and a few cells depicted nuclear staining also. Only a subset of connective tissue cells in lamina propria depicted positivity (cytoplasmic) for this protein.
Western Blot: Caspase-1 Antibody (14F468) [NB100-56565] - P. gingivalis and its OMVs differentially induce inflammasome signaling and pyroptosis in murine macrophages.
