Western Blot: Glutathione S-Transferase pi 1/GSTP1 Antibody (11D5) [NBP2-76868] - Western blot analysis of GST3 on A549 cell lysate. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody was used at a 1:500 dilution in 5% BSA at room temperature for 2 hours. Goat Anti-Mouse IgG - HRP Secondary Antibody (HA1006) at 1:5,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: Glutathione S-Transferase pi 1/GSTP1 Antibody (11D5) [NBP2-76868] - Immunohistochemical analysis of paraffin-embedded mouse lung tissue using anti-GST3 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the antibody at 1/200 dilution, for 30 minutes at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chrogen. Counter stained with hematoxylin and mounted with DPX.

Flow Cytometry: Glutathione S-Transferase pi 1/GSTP1 Antibody (11D5) [NBP2-76868] - Flow cytometric analysis of GST3 was done on HCT116 cells. The cells were fixed, permeabilized and stained with GST3 antibody at 1/100 dilution (red) compared with an unlabelled control (cells without incubation with primary antibody; black). After incubation of the primary antibody on room temperature for an hour, the cells was stained with a Alexa Fluor 488-conjugated goat anti-mouse IgG Secondary antibody at 1/500 dilution for 30 minutes.

DAB was used as the chrogen. Counter stained with hematoxylin and mounted with DPX.

Background

GSTP1 (glutathione-S-transferase, pi 1), also called GST3/DFN7, which is located on chromosome 11q13 , is a family of enzymes that play an important role in detoxification by catalyzing the conjugation of many hydrophobic and electrophilic compounds with reduced glutathione. GSTP1 act like a tumor suppressor gene, which when inactivated leads to tumor growth, and the -class glutathione S-transferase is commonly inactivated by somatic CpGisland hypermethylation in cancers of the prostate, liver, and breast. Methylation of regulatory sequences at the GSTP1 gene locus is found in the vast majority (>90%) of prostate carcinomas and is associated with transcriptional down-regulation.
