Background

This gene encodes a phosphorylated protein that is a member of a Skp1-Cullin-F-box-like complex. The protein is essential for normal development of the vasculature and mutations in this gene have been associated with glomuvenous malformations, also called glomangiomas. Alternatively spliced variants that encode different protein isoforms have been described but the full-length nature of only one has been determined. [provided by RefSeq]

Immunocytochemistry/Immunofluorescence: APC Antibody [NB100-91662] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-APC Antibody at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
Flow Cytometry: APC Antibody [NB100-91662] - An intracellular stain was performed on HeLa cells with APC Antibody NB100-91662AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Simple Western lane view shows lysates of LoVo human colorectal adenocarcinoma cell line, loaded at 0.2 mg/mL. A specific band was detected for DPPIV/CD26 at approximately 168 kDa (as indicated) using 2 µg/mL of Goat Anti-Human DPPIV/CD26 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1180) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.
DPPIV/CD26 was detected in immersion fixed paraffin-embedded sections of human psoriatic skin using Goat Anti-Human DPPIV/CD26 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1180) at 1 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Specific staining was localized to keratinocytes. Knockout Validated: c-Fos Antibody (2H2) [NBP2-50037] - Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and c-Fox knockout (KO) HeLa cell line. PVDF membrane was probed with 1:1000 of Mouse Anti-Human c-Fox Monoclonal Antibody (Catalog # NBP2-50037) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog #HAF018). Specific band was detected for c-Fox at approximately 52 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.
Immunocytochemistry/Immunofluorescence: c-Fos Antibody (2H2) [NBP2-50037] - Section of rat hippocampus stained with mouse monoclonal antibody to c-FOS NBP2-50037 in red and counterstained with rabbit polyclonal antibody to FOX3/NeuN. DAPI reveals nuclei of neurons and glia in blue. The hippocampal neurons stain green for FOX3/NeuN and a few also are expressing c-FOS, and so appear orange. These cells were spontaneously active at the time the animal was sacrificed.
Immunocytochemistry/Immunofluorescence: c-Fos Antibody (2H2) [NBP2-50037] - Left: NBP2-50037 staining (green) in HeLa cells, which were treated with serum-starvation for 36 hrs, followed by 2 hrs 20% FBS stimulation (bottom), or PBS treatment (top). Green c-Fos staining only localizes in the nuclei of stimulated cells, but not in un-stimulated cells. Cells are counter-stained with chicken pAb against Vimentin (NB300-223, red). Recombinant Human TNF-alpha (Catalog # 210‑TA) induces cytotoxicity in the L-929 mouse fibroblast cell line in the presence of the metabolic inhibitor actinomycin D. The ED50 for this effect is 25‑100 pg/mL.
