Background
FXR2 (Fragile X relative protein 2) belongs to FMR1 family/fragile X mental retardation proteins which can bind to RNA and associate with polyribosomes. All three members of FMR1 proteins i.e. FMRP, FXR1, and FXR2 contains RNA-binding domains and RGG boxes, and are found predominantly in cytoplasm where they can associate with ribosomes for implicating translation or mRNA stability. FXR2 has been shown to interact with FMR1, FXR1, CYFIP2 and TDRD3. It is ubiquitously expressed during embryonic development and its tissue distribution overlaps with FMRP in adult stages. However, interestingly, FXR2 knockout mice display learning impairments and changes in synaptic plasticity that are somewhat distinct from those of FMRP-deficient mice, indicating that FXR2 may also regulate adult hippocampal neurogenesis via mechanisms distinct from FMRP. FXR2 deficiency leads to altered stem cell proliferation and differentiation specifically in dentate gyrus and not in subventricular zone which has been suggested as a key component of mechanism regulating differential neurogenic processes in these two adult germinal zones.
