Western Blot: Dopa Decarboxylase/DDC Antibody (JA53-16) [NBP2-66806] - Analysis of Dopa Decarboxylase/DDC on HepG2 cell lysates with Rabbit anti-Dopa Decarboxylase/DDC antibody at 1/500 dilution. Lysates/proteins at 10 ug/Lane. Predicted band size: 53 kDa Observed band size: 55 kDa Exposure time: 2 minutes; 10% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:200,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry-Paraffin: Dopa Decarboxylase/DDC Antibody (JA53-16) [NBP2-66806] - Analysis of paraffin-embedded human liver tissue with Rabbit anti-DOPA Decarboxylase antibody washed with ddH2O and PBS, and then probed with the primary antibody at 1/400 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

DOPA Decarboxylase (aromatic L-amino acid decarboxylase, AADC; DDC) catalyzes the second reaction in the biosynthesis of catecholamines and serotonin (Waymire and Haycock, 2002; Berry et al., 1996; Haycock et al., 2003). It is also involved in the biosynthesis of trace amines. DDC antibodies can therefore be used as markers for dopaminergic, noradrenergic and serotonergic neurons in a variety of applications including depression, schizophrenia, Parkinson's disease and drug abuse (Kish et al., 2001; Zhu et al., 2000; Zhu et al., 1999).
